A nucleoside is composed of a nitrogenous base and a five-carbon sugar (ribose or deoxyribose), which can be thought as a nucleotide without a phosphate group. The nitrogenous base is bond to the pentose via a beta-glycosidic linkage. Examples of nucleosides include cytidine, uridine, adenosine, guanosine, thymidine and inosine. Nucleosides are the basic building blocks of nucleic acids (DNA and RNA) that play a significant role in genetic information expression.

Application

  • Nucleoside analogues are used as antiviral or anticancer agents in medicine.
  • Several analogues of the sugar backbone exist in molecular biology, which are more stable alternative nucleoside/nucleotide analogues including LNA, morpholino, PNA.
  • Dideoxynucleotides are used in sequencing. These nucleotides possess the non-canonical sugar dideoxyribose, which lacks 3’ hydroxyl group (which accepts the phosphate), therefore it cannot bond with the next base and terminates the chain.
