Metoprolol Tartrate (Fig. 1b) bis[(2RS)-1-[4-(2-methoxyethyl)phenoxy]-3-[(1-methylethyl)amino]propane-2-ol](2R,3R)-2,3-dihydroxybutanedioate is a prototype b-1 anti-adrenergic drug which reduces chest pain and lowers the blood pressure. Metoprolol free base is highly lipophilic; therefore, it is available either as succinate, fumarate or tartrate salts, which show different solubility profiles. Metoprolol Tartrate is a salt consisting of a racemic mixture of optical isomers of the base and dextrotartaric acid in a 2:1 ratio, and this form will be discussed in further detail as this was selected for experimental work.

The objective of this study was therefore to determine the impact of ethylcellulose molecular weight (as measured by viscosity) variation within the manufacturer’s specification on the release of two model drugs metoprolol tartrate and acetaminophen. Metoprolol tartrate was selected to represent a model drug with high aqueous solubility, whereas acetaminophen as a model drug with low aqueous solubility. Drug layered sugar spheres were coated with different viscosity grades of ethylcellulose at varying coating weight gains, and their drug release profiles were determined.

The structure of metoprolol tartrate has been investigated by a meticulous NMR study. The 1H NMR signals were assigned unambiguously on the basis of two-dimensional HSQC and they were confirmed by 1H–1H COSY and HMBC. The spectrum on Fig. 4 obtained experimentally is quite similar to the one previously reported with very minor differences in the position of some hydrogen signals since it was obtained in CDCl3. A worth mentioning difference is that b-H and b0-H are chemically equivalent and gave a singlet signal at 4.39 in CDCl3 while they gave the same singlet at 3.99 in DMSO. The total assignment of 1H NMR signals for metoprolol tartrate is given in Table 1.
