1.[Comparative evaluation of methacycline hydrochloride capsules and tablets according to their dissolubility and rate of solution].

Desintegration and dissolution of capsules and tablets of methacycline hydrochloride were studied. The study on solubility of methacycline hydrochliride capsules filled with methacycline granulate or powder according to the same formula showed that the rate of the antibiotic liberation from the capsules filled with the powder decreased during storage while that from the capsules filled with the granulate did not change. Investigation of the effect of the mass packing value in a drop on the antibiotic liberation from the capsules showed that an increase in the packing coefficient above 1.38 resulted in a marked decrease in the rate of methacycline liberation from the capsules filled with the granulate. No correlation between desintegration and dissolution of methacycline capsules and tablets was found.

Drug induced esophageal ulcers are more frequent than supposed. Four ulcerative lesions of the esophagus, two after ingestion of doxycycline-hydrochloride, one after metacycline-hydrochloride, and one after emeproniumbromide were detected at the department of Internal Medicine of a medium-sized standard-hospital during the last six years. Symptoms develop suddenly after taking the drug with minimum fluid, usually before going to bed. Retrosternal burning, odynophagia and dysphagia are pathognomonic features. The cause of ulceration is discussed to be a matter of the drug remaining in the esophagus and of its local cytolytic and corrosive effect. Cytomegalo and herpes-simplex-infections can be excluded as etiologic factor of such ulcers. The clinical diagnosis is verified by endoscopy only.

A quick colorimetric method is reported for the determination of tetracycline derivatives such as oxytetracycline hydrochloride (OTCH), chlortetracycline hydrochloride (CTCH), methacycline hydrochloride (MCH), and doxycycline hydrochloride (DCH). The method involves complexation of the above derivatives with cupric chloride in alkaline medium. The yellowish green copper complexes of OTCH, CTCH, MCH, and DCH show maximum absorbance at 395, 410, 400, and 400 nm, respectively. The color intensity obeys Beer's law in the concentration range of 0-20 micrograms/mL.
