1.PCL/PHBV microparticles as innovative carriers for oral controlled release of manidipine dihydrochloride.
Barboza FM1, Machado WM2, Olchanheski Junior LR2, Padilha de Paula J1, Zawadzki SF3, Fernandes D2, Farago PV1. ScientificWorldJournal. 2014 Jan 16;2014:268107. doi: 10.1155/2014/268107. eCollection 2014.
Microparticles of poly(ε-caprolactone) (PCL) and poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) containing manidipine dihydrochloride (MAN) were successfully prepared by the simple emulsion/solvent evaporation method. All formulations showed loading efficiency rates greater than 80% and average particle size less than 8 μm. Formulations had spherical shape with smooth and porous surface for PCL and PHBV, respectively. According to Fourier-transform infrared spectroscopy, initial components were not chemically modified during microencapsulation. X-ray diffraction patterns and differential scanning calorimetry demonstrated that this process led to drug amorphization. In vitro dissolution studies showed that all microparticles prolonged MAN release, mainly which one obtained using PCL that contained 5% of drug loaded (PCL-M5). Animal studies demonstrated that formulation PCL-M5 was able to keep the variation of mean arterial pressure after phenylephrine administration up to 24 hours.
2.[Cost-effectiveness analysis of fixed dose antihypertensive drugs].
García Ruiz AJ1, Divisón Garrote JA, García-Agua Soler N, Morata García de la Puerta F, Montesinos Gálvez AC, Avila Lachica L. Semergen. 2013 Mar;39(2):77-84. doi: 10.1016/j.semerg.2012.05.011. Epub 2012 Sep 5.
OBJECTIVE: The aim of this study is to compare the efficiency of different fixed-dose combinations of renin-angiotensin-aldosterone system (RAAS) blockers and calcium channel blockers, to use it as a guide to assist the rational prescribing in antihypertensive therapy.
3.Effects of manidipine vs. amlodipine on intrarenal haemodynamics in patients with arterial hypertension.
Ott C1, Schneider MP, Raff U, Ritt M, Striepe K, Alberici M, Schmieder RE. Br J Clin Pharmacol. 2013 Jan;75(1):129-35. doi: 10.1111/j.1365-2125.2012.04336.x.
AIMS: Intraglomerular pressure is one of the main drivers of progression of renal failure. Experimental data suggest that there are important differences between calcium channel blockers (CCBs) in their renal haemodynamic effects: manidipine reduces, whereas amlodipine increases intraglomerular pressure. The aim of this study was to investigate the effects of manidipine and amlodipine treatment on intragomerular pressure (P(glom)) in patients with mild to moderate essential hypertension.
4.First-order derivative UV spectrophotometric method for simultaneous measurement of delapril and manidipine in tablets.
Todeschini V1, Barden AT, Sfair LL, da Silva Sangoi M, Volpato NM. Acta Chim Slov. 2013;60(2):335-42.
A first-order derivative spectrophotometric (1D-UV) method was developed and validated for simultaneous determination of delapril (DEL) and manidipine (MAN) in tablets. The 1D-UV spectra were obtained using change lambda = 4.0 nm and wavelength set at 228 nm for DEL and 246 nm for MAN. The method was validated in accordance with the ICH requirements, involving the specificity, linearity, precision, accuracy, robustness and limits of detection and quantitation. The method showed high specificity in the presence of two drugs and formulation excipients and was linear over the concentration range of 18-54 microg mL(-1) (r2 = 0.9994) for DEL and 6-18 microg mL(-1) (r2 = 0.9981) for MAN with adequate results for the precision (< or = 1.47%) and accuracy (98.98% for DEL and 100.50% for MAN). Moreover, the method proved to be robust by a Plackett-Burman experimental design evaluation. The proposed 'D-UV method was successfully applied for simultaneous analysis of DEL and MAN in tablets and can be used as alternative green method to separation techniques.
