Multiple phenolic compounds in the extract of Erigeron breviscapus synergistically contribute to the neurovascular protective effects. We conducted a phase I and pharmacokinetic study with the phenolic compound-enriched product extracted from Erigeron breviscapus, Erigerontis hydroxybenzenes injection (EHI), in healthy Chinese volunteers. A randomized, open-label, single-center, double-arm, dose-escalation study of EHI was conducted. The tolerability of intravenously EHI administrated in single- or multiple-dose (once daily for 7 days) was studied in 40 healthy Chinese volunteers and the pharmacokinetics of EHI was studied in additional 10 volunteers. The tolerated dose of intravenous infusion of EHI in healthy Chinese volunteers was 6 vials (equivalent to 90 mg bioactive phenolic compounds). The main limitations to dose escalation of EHI were transit changes in electrocardiogram and mild, transit increase in alanine aminotransferase. After intravenous administration of EHI, the average systemic clearance of multiple phenolic compounds of scutellarin, 1,3-dicaffeoylquinic acid, 3,5-dicaffeoylquinic acid, and 3,4-dicaffeoylquinic acid were 131, 29, 262, 112 L/h for male volunteers and 202, 28, 252, 117 L/h for female volunteers.
2.Isolation and identification of antioxidant compounds from Ligularia fischeri.

The plant Ligularia fischeri var. spiciformis Nakai, a well-known edible medicinal herb in Korea, has been used to treat maladies such as jaundice, scarlet fever, rheumatoid arthritis, and hepatic function failure. In this research, 4 major antioxidant compounds were detected from this plant's leaves using an on-line high-performance liquid chromatography (HPLC)-ABTS screening system, which can determine the antioxidant activity based on a decrease in absorbance at 734 nm after postcolumn reaction of HPLC-separated antioxidants with the 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) radicals (ABTS(*)). In order to isolate these active compounds, a preparative HPLC was applied and their chemical structures were identified as 5-O-caffeoylquinic acid (5-CQA), 3,4-di-O-caffeoylquinic acid (3,4-DCQA), 3,5-di-O-caffeoylquinic acid (3,5-DCQA), and 4,5-di-O-caffeoylquinic acid (4,5-DCQA) by ESI/MS(n) and (1)H NMR. These 4 isomers comprised over 10% of the dried leaves, with 3,5-DCQA being the most abundant compound. The radical scavenging activity of each isomer was also evaluated simultaneously through the on-line HPLC-ABTS method, which showed 94% antioxidant activity of the ethanol extract derived from caffeoylquinic acids.

BACKGROUND: ;Ilex paraguariensis St. Hilaire (Aquifoliaceae) ("Mate" or "Yerba mate") is one of the most commercialized plants of South America which grows naturally in NE Argentina, Uruguay, SE Brazil and E Paraguay, where it is also cultivated. It is used to prepare a tea-like beverage (infusions or decoctions) appreciated for its peculiar flavor, stimulation and nutritional properties. Ilex brevicuspis Reisseck grows in the same habitat and is widely used as a substitute or adulterant of Ilex paraguariensis. In a previous work, methylxanthines (caffeine, theobromine and theophylline) were not detected in it by HPLC.;AIM OF THE STUDY: ;This study was undertaken in order to isolate, identify and quantify the polyphenolic compounds (caffeoyl derivatives and flavonoids) and to investigate some of the pharmacological activities of I. brevicuspis, related with the traditional use of the "Mate" (choleretic, intestinal propulsion and antioxidant activities). Acute toxicity was also investigated.;METHODS: ;Decoctions, like extracts, were prepared in order to compare the results with preparations commonly used by the local people. For the phytochemical analysis, the extracts were analyzed by HPLC with a diode array detector.
