1.Preparation of a β-Cyclodextrin-Based Open-Tubular Capillary Electrochromatography Column and Application for Enantioseparations of Ten Basic Drugs.
Fang L;Yu J;Jiang Z;Guo X PLoS One. 2016 Jan 15;11(1):e0146292. doi: 10.1371/journal.pone.0146292. eCollection 2016.
An open-tubular capillary electrochromatography column was prepared by chemically immobilized β-cyclodextrin modified gold nanoparticles onto new surface with the prederivatization of (3-mercaptopropyl)-trimethoxysilane. The synthesized nanoparticles and the prepared column were characterized by transmission electron microscopy, scanning electron microscopy, infrared spectroscopy and ultraviolet visible spectroscopy. When the column was employed as the chiral stationary phase, no enantioselectivity was observed for ten model basic drugs. So β-cyclodextrin was added to the background electrolyte as chiral additive to expect a possible synergistic effect occurring and resulting in a better separation. Fortunately, significant improvement in enantioselectivity was obtained for ten pairs of drug enantiomers. Then, the effects of β-cyclodextrin concentration and background electrolyte pH on the chiral separation were investigated. With the developed separation mode, all the enantiomers (except for venlafaxine) were baseline separated in resolutions of 4.49, 1.68, 1.88, 1.57, 2.52, 2.33, 3.24, 1.63 and 3.90 for zopiclone, chlorphenamine maleate, brompheniramine maleate, dioxopromethazine hydrochloride, carvedilol, homatropine hydrobromide, homatropine methylbromide, venlafaxine, sibutramine hydrochloride and terbutaline sulfate, respectively.
2.Use of topical nonsteroidal anti-inflammatory drugs in excimer laser photorefractive keratectomy.
Arshinoff S;D'Addario D;Sadler C;Bilotta R;Johnson TM J Cataract Refract Surg. 1994 Mar;20 Suppl:216-22.
Nonsteroidal anti-inflammatory drugs (NSAIDs) produce potent analgesic, antipyretic, and anti-inflammatory effects. We studied postoperative pain in 97 consecutive patients having photorefractive keratectomy (PRK) by an excimer laser with different topical NSAID protocols. Treatment with topical homatropine hydrobromide, either diclofenac sodium (Voltaren Ophthalmic) or ketorolac tromethamine (Acular), and a soft contact lens was most effective in achieving post-PRK analgesia. We also studied post-PRK myopic regression in 68 consecutive patients and found that flurbiprofen sodium (Ocufen), when added to topical steroid protocols, significantly reduced myopic regression for one year postoperatively more than steroids alone or steroids and diclofenac sodium. Diclofenac, used with topical steroids, had less of an additive effect on myopic regression than did flurbiprofen. Topical NSAIDs are useful adjuncts to PRK therapy, both to eliminate postoperative pain and to control post-PRK myopic regression.
3.Combined use of ionic liquid and β-CD for enantioseparation of 12 pharmaceuticals using CE.
Zuo L;Meng H;Wu J;Jiang Z;Xu S;Guo X J Sep Sci. 2013 Feb;36(3):517-23. doi: 10.1002/jssc.201200824. Epub 2013 Jan 10.
In this study, the enantioseparation of zopiclone, repaglinide, chlorphenamine maleate, brompheniramine maleate, dioxopromethazine hydrochloride, promethazine hydrochloride, liarozole, carvedilol, homatropine hydrobromide, homatropine methylbromide, venlafaxine, and sibutramine hydrochloride has been investigated using β-CD in combination with a chiral ionic liquid (IL), 1-ethyl-3-methylimidazolium-L-lactate. The influence of the type of IL and its concentration, BGE pH, and chain length of the IL cations on the resolution are discussed. Finally, the proposed method was successfully applied for the chiral impurity determination of eszopiclone in pharmaceutical tablets after validation with respect to accuracy and precision, linearity range, selectivity, repeatability, LOD and LOQ. It is assessed that the chiral impurity determination in the commercial tables is fewer than 0.1%.
