Description:
Escitalopram Oxalate also known by the brand names Lexapro and Cipralex among others, is an antidepressant of the selective serotonin reuptake inhibitor (SSRI) class. It is approved by the U.S. Food and Drug Administration (FDA) for the treatment of adults and children over 12 years of age with major depressive disorder (MDD) and generalized anxiety disorder (GAD).

Half of the context-conditioned (CC-Escit) and half of the non-conditioned rats (NCC-Escit) received a single injection of escitalopram (escitalopram oxalate, 10 mg/kg, from a solution of 4 mg per ml of 0.9 % saline) 1 h before test 3. The two remaining groups (CC-NaCl and NCC-NaCl) received 1ml of 0.9 % saline. No injections were given prior to test 1, test 2 or training.

Escitalopram oxalate (CIT), S-(+)-1-[3-(dimethyla-mino) propyl]-1-(4-fluorophenyl)-1,3-dihydro-5-isobenzofuran-carbonitrile oxalate, is a selective serotonin reuptake inhibitor. It is an active S (+)-enantiomer present in the racemate citalopram. As a new SSRI, CIT shows greater efficacy, better tolerance and less adverse reactions than citalopram. Recent studies about SSRIs have focused on their neuroprotective effect on ICD, especially on acute ICD. However, the use of SSRIs in chronic hypoperfusion has been seldom reported, much less for the protective effect of CIT on chronic hypoperfusion.

The free escitalopram was extracted by ethyl acetate from escitalopram oxalate (ESP-OA) solution which was adjusted to pH 11 by sodium hydroxide (4%, w/v). It was identified by differential scanning calorimetry (DSC) and Fourier transform infrared (FTIR). Acetone was used as a solvent for preparing ion-pair complexes. The mixing solutions with free escitalopram and equimolar amounts of organic acids were stirred for approximately 2 h, then concentrated under vacuum, and the ESP-acid complexes were obtained and identified by DSC and 13C NMR.

A literature survey revealed that assay of escitalopram oxalate either pure or in dosage forms is not official in any pharmacopoeia and therefore requires much investigation. Several analytical methods have been reported for the determination of escitalopram in biological fluids and pharmaceutical formulations; these include liquid chromatography coupled with mass spectrometry, HPLC, chiral liquid chromatography, and capillary electrophoresis. In these methods escitalopram is analyzed either alone or in combination with drugs other than clonazepam. The United States Pharmacopoeia describes an HPLC method for deter- mination of clonazepam in tablet formulations. Analytical methods reported for the determination of clonazepam in biological fluids, as the bulk drug, and in pharmaceutical formulations include HPLC, spectrophotometry, fluorimetry, potentiometry, a GC–MS stability-indicating method (determination of the drug in the presence of its metabolites or impurities), Fourier-transform Raman, and infrared spectrophotometry. In these methods clonazepam is analyzed either alone or in combination with drugs other than escitalopram oxalate.
