1.Dopamine Increases a Value-Independent Gambling Propensity.
Rigoli F1, Rutledge RB1,2, Chew B1, Ousdal OT1,3, Dayan P4, Dolan RJ1,2. Neuropsychopharmacology. 2016 May 6. doi: 10.1038/npp.2016.68. [Epub ahead of print]
While the impact of dopamine on reward learning is well documented, its influence on other aspects of behaviour remains the subject of much ongoing work. Dopaminergic drugs are known to increase risk-taking behaviour, but the underlying mechanisms for this effect are not clear. We probed dopamine's role by examining the effect of its precursor L-DOPA on the choices of healthy human participants in an experimental paradigm which allowed particular components of risk to be distinguished. We show that choice behaviour depended on a baseline (ie, value-independent) gambling propensity, a gambling preference scaling with the amount/variance, and a value normalization factor. Boosting dopamine levels specifically increased just the value-independent baseline gambling propensity, leaving the other components unaffected. Our results indicate that the influence of dopamine on choice behaviour involves a specific modulation of the attractiveness of risky options-a finding with implications for understanding a range of reward-related psychopathologies including addiction.
2.The crystal structure of human dopamine β-hydroxylase at 2.9 Å resolution.
Vendelboe TV1, Harris P1, Zhao Y2, Walter TS2, Harlos K2, El Omari K2, Christensen HE1. Sci Adv. 2016 Apr 8;2(4):e1500980. doi: 10.1126/sciadv.1500980. eCollection 2016.
The norepinephrine pathway is believed to modulate behavioral and physiological processes, such as mood, overall arousal, and attention. Furthermore, abnormalities in the pathway have been linked to numerous diseases, for example hypertension, depression, anxiety, Parkinson's disease, schizophrenia, Alzheimer's disease, attention deficit hyperactivity disorder, and cocaine dependence. We report the crystal structure of human dopamine β-hydroxylase, which is the enzyme converting dopamine to norepinephrine. The structure of the DOMON (dopamine β-monooxygenase N-terminal) domain, also found in >1600 other proteins, reveals a possible metal-binding site and a ligand-binding pocket. The catalytic core structure shows two different conformations: an open active site, as also seen in another member of this enzyme family [the peptidylglycine α-hydroxylating (and α-amidating) monooxygenase], and a closed active site structure, in which the two copper-binding sites are only 4 to 5 Å apart, in what might be a coupled binuclear copper site.
3.Simple and Sensitive Colorimetric Detection of Dopamine Based on Assembly of Cyclodextrin-Modified Au Nanoparticles.
Wen D1, Liu W1, Herrmann AK1, Haubold D1, Holzschuh M2, Simon F2, Eychmüller A1. Small. 2016 May;12(18):2439-42. doi: 10.1002/smll.201503874. Epub 2016 Mar 22.
A controlled assembly of natural beta-cyclodextrin modified Au NPs mediated by dopamine is demonstrated. Furthermore, a simple and sensitive colorimetric detection for dopamine is established by the concentration-dependent assembly.
4.Treatment of clinically non-functioning pituitary adenomas with dopamine agonists.

OBJECTIVE: Clinically nonfunctioning pituitary adenoma (NFPA), remain the only pituitary tumor subtype for which no effective medical therapy is available or recommended. We evaluated dopamine agonist (DA) therapy for preventing growth of post-surgical pituitary tumor remnants.
