1.HPLC method for identification and quantification of three active substances in a dermatological preparation--Viosept ointment.
Podolska M, Kulik A, Białecka W, Kwiatkowska-Puchniarz B, Mazurek A. Acta Pol Pharm. 2014 Sep-Oct;71(5):709-19.
The study was aimed at developing a HPLC method to identify and quantify domiphen bromide, tripelennamine hydrochloride and clioquinol in Viosept ointment. The tested substances were successfully separated using Inertsil ODS-3 (250 x 4.6 mm, 5 μm) as a stationary phase and a gradient elution. Detection at 310 nm wavelength was applied for tripelennamine hydrochloride and clioquinol, and at 215 nm wavelength for domiphen bromide. Methods of extraction of the tested substances were developed: domiphen bromide and clioquinol were extracted with acetone from heated solutions, and tripelennamine hydrochloride was extracted in a hexane-water system. Validation procedure confirmed the method to be sufficiently selective, precise and accurate. Correlation coefficients of calibration curves pointed out that they were linear within the examined concentration range.
2.Development of a novel adenovirus purification process utilizing selective precipitation of cellular DNA.
Goerke AR1, To BC, Lee AL, Sagar SL, Konz JO. Biotechnol Bioeng. 2005 Jul 5;91(1):12-21.
The use of recombinant adenoviral vectors for vaccination and gene therapy requires the development of purification processes that are cost-effective, scalable, and capable of robust host cell DNA clearance. An adenovirus purification process was developed which incorporates selective precipitation of host cell DNA, enabling a reduction in the use of costly nucleases and chromatographic resins while substantially improving DNA and protein clearance capabilities. In this work, three cationic detergents were evaluated for their capacity to selectively precipitate DNA from adenovirus-containing cell lysate. Parameters including pH, sodium chloride concentration, nonionic surfactant concentration, and cell density were investigated during development of the precipitation step. In a novel application, the cationic detergent domiphen bromide was found to have superior selectivity for host cell DNA. In addition, domiphen bromide-induced precipitation of adenovirus was shown to be reversible, which reduces the importance of mixing.
3.Characterization of micelle formation of dodecyldimethyl-N-2-phenoxyethylammonium bromide in aqueous solution.
Khatua D1, Gupta A, Dey J. J Colloid Interface Sci. 2006 Jun 1;298(1):451-6. Epub 2006 Jan 18.
Aggregation behavior of dodecyldimethyl-N-2-phenoxyethylammonium bromide commonly called domiphen bromide (DB) was studied in aqueous solution. The Krafft temperature of the surfactant was measured. The surfactant has been shown to form micellar structures in a wide concentration range. The critical micelle concentration was determined by surface tension, conductivity, and fluorescence methods. The conductivity data were also employed to determine the degree of surfactant counterion dissociation. The changes in Gibb's free energy, enthalpy, and entropy of the micellization process were determined at different temperature. The steady-state fluorescence quenching measurements with pyrene and N-phenyl-1-naphthylamine as fluorescence probes were performed to obtain micellar aggregation number. The results were compared with those of dodecyltrimethylammonium bromide (DTAB) surfactant. The micelle formation is energetically more favored in DB compared to that in DTAB.
4.Inhibition of HERG potassium channels by domiphen bromide and didecyl dimethylammonium bromide.
Long Y1, Chen W2, Lin Z1, Sun H3, Xia M3, Zheng W3, Li Z4. Eur J Pharmacol. 2014 Aug 15;737:202-9. doi: 10.1016/j.ejphar.2014.05.002. Epub 2014 May 15.
Domiphen bromide and didecyl dimethylammonium bromide were widely used environmental chemicals with potent activity on blockade of human ether-a-go-go related gene (HERG) channels. But the mechanism of their action is not clear. The kinetics of block of HERG channels by domiphen bromide and didecyl dimethylammonium bromide was studied in order to characterize the inhibition of HERG currents by these quaternary ammonium compounds (QACs). Domiphen bromide and didecyl dimethylammonium bromide inhibited HERG channel currents in a dose-dependent manner with IC50 values of 9nM and 5nM, respectively. Block of HERG channel by domiphen bromide and didecyl dimethylammonium bromide was voltage-dependent and use-dependent. Domiphen bromide and didecyl dimethylammonium bromide caused substantial negative shift of the activation curves, accelerated activated process, but had no effects on the deactivation and reactivation processes. The docking models implied that these two compounds bound to PAS domain of HERG channels and inhibited its function.
