Amitriptyline.HCl (AMI) and clomipramine.HCl (CMI) belong to the category of tricyclic antidepressant drugs (TCAs) and are widely used to treat various kinds of depression. Amitriptyline-HCl, (Fig. 1a) 3-(10,11-dihydro-5H-dibenzo [a,d] cycloheptene-5-ylidene)-N,N-dimethyl propan-1-amine hydrochloride, is useful in the treatment of involutional melancholia, anxiety and manic depression. Clomipramine-HCl, (Fig. 1b) 3-(3-chloro-10,11-dihydro-5H-dibenzo[b,f]azepin-5-yl)-N,N-dimethylpropan-1-amine hydrochloride, is mainly used for the treatment and relief of obsessive and compulsive disorders as well as in depression and other emotional disturbances. Side effects of AMI and CMI are essentially those of dibenzazepines which include drowsiness, hypothermia, arrhythmic abnormalities, congestive heart failure and death in cases of an overdose. The majority of the TCAs act primarily by inhibiting serotoninnorepinephrine reuptake in central nervous system which results in an elevation of the synaptic concentrations of these neurotransmitters, and therefore an enhancement of neurotransmission. In spite of new atypical drugs, such as those of the SSRI group (fluoxetine, fluvoxamine, etc.), AMI and CMI are still the reference compounds in the treatment of many psychiatric disorders. However, these are considered to have many undesirable side effects and overdosage may cause adverse reactions.

Clomipramine hydrochloride is the generic name of 3-chloro-10,11-dihydro-N,N-dimethyl-5H-dibenz[b,f]azepine-5-propanamine hydrochloride. It is a white to off-white powder, very soluble in water. Its action and uses are similar to other tricyclic antidepressants. It is used to treat depression, migraine headaches, panic disorders, bedwetting and with other drugs to manage chronic/severe pain. Many analytical techniques have been employed for the determination of clomipramine–HCl in pure and dosage forms. However, relatively less attention has been paid to the development of spectrophotometric methods for the assay of clomipramine–HCl in pure and dosage forms. However, such spectrophotometric methods suffer from poor detection limits (up to 50 mg/L). Extraction and determination of clomipramine in urine samples using dispersive liquid–liquid micro extraction followed by high performance liquid chromatog raphy (HPLS) is reported [4]. A similar method based on simultaneous determination of clomipramine and its desmethyl and hydroxy metabolites in plasma of patients by HPLC after solid phase extraction (SPE) is present. Another method based on multicommutated flow system for the chemiluminescent determination of clomipramine in pharmaceutical preparations is reported. A comparative SPE study for the simultaneous determination of fluoxetine, amitriptyline, nortriptyline, trimipramine, maprotiline, clomipramine, and trazodone in whole blood by capillary gas–liquid chromatography with nitrogen–phosphorus detection was described by Martinez.
