1.Pharmacokinetic comparison of seven major bioactive components in normal and depression model rats after oral administration of Baihe Zhimu decoction by liquid chromatography-tandem mass spectrometry.

A simple and rapid liquid chromatography-tandem mass spectrometry method was firstly developed for simultaneous quantification of neomangiferin, mangiferin, regaloside A, regaloside I, timosaponin BII, anemarsaponin E and timosaponin AIII in rat plasma after oral administration of Baihe Zhimu decoction, which plays an important role for the treatment of depression. The plasma samples were pretreated by a one-step direct protein precipitation with methanol. Separation of the seven components and scutellarin (IS) from endogenous components with high selectivity and sensitivity (LLOQ, 0.1-1.0ng/mL) was achieved within 10min using Poroshell 120 EC-C;18; column (150mm×3.0mm, 2.7μm). A gradient mobile phase consisting of acetonitrile and water (containing 5mM ammonium acetate) was applied at a flow rate of 0.4mL/min. Detection and measurement were performed on an AB Sciex QTRAP;®; 5500 mass spectrometer in multiple reactions monitoring mode. The intra- and inter-day precisions were all within 15% and the accuracies were in the range of -10.4% to 14.5%. The recovery ranged from 90.8 to 113.8%. The validated method was successfully applied to pharmacokinetic study of the seven components in normal and chronic unpredicted mild stress-induced depression model rats.
2.[HPLC-PDA-CAD Fingerprints of Salt Anemarrhenae Rhizoma].

OBJECTIVE: ;To establish the HPLC-PDA-CAD fingerprints and to determine its seven main constituents so as to provide a reliable evidence for the scientific evaluation and quality control of salt Anemarrhenae Rhizoma.;METHODS: ;The chromatographic fingerprint was obtained with Thermo Hypersil C18 column (250 mm x 4.6 mm, 5 μm) and gradient eluted with acetonitrile and water; The CAD parameters were pressure of 241. 3 kPa, filter of high and range of 200 pA. The detection wavelength of PDA was set at 258 nm.;RESULTS: ;The common mode of HPLC-PDA-CAD fingerprint of salt Anemarrhenae Rhizoma was set up. There were 5 PDA and 12 CAD common peaks in the fingerprints. Timosaponin B II, anemarsaponin E, timosaponin B III, timosaponin A III, neomangiferin, mangiferin and baohuoside I were identified in fingerprints and determined.;CONCLUSION: ;The established HPLC-PDA-CAD fingerprint method is accurate, reliable, and has a good reproducibility and precision, which can be used for the quality control of salt Anemarrhenae Rhizoma.
3.Effect of six steroidal saponins isolated from anemarrhenae rhizoma on platelet aggregation and hemolysis in human blood.

Six steroidal saponins were isolated from Anemarrhena asphodeloides Bunge (Liliaceae), a traditional chinese medicine, and named anemarrhenasaponin I (An-I), anemarrhenasaponin Ia (An-Ia), timosaponin B-I (TB-I), timosaponin B-II (TB-II), timosaponin B-III (TB-III), and timosaponin A-III (TA-III). The effects of these six compounds on platelet aggregation and hemolysis in human blood were studied. All these compounds provoked remarkable inhibiting effect on platelet aggregation, and activated partial thromboplastin times (APTT) are sensitive to the presence of these six compounds. Using an in vitro system, APTT was delayed with the increment of the concentrations of these six compounds. In these six compounds, only timosaponin A-III appeared a strong effect on hemolysis, and anemarrhenasaponin Ia had a slight effect on hemolysis, other had no effect on hemolysis. These results suggested that these steroidal saponins isolated from Anemarrhena asphodeloides Bunge (Liliaceae) might be used as a novel antithrombotic therapeutic agents in post-myocardial infarction.
