1. Isolation and characterization of related impurities in andrographolide sodium bisulphite injection

Andrographolide sodium bisulphite (ASB), a water-soluble sulfonate of andrographolide isolated as the major bioactive diterpenoid lactone of Andrographis paniculata Nees (Acantha-ceae), has been used clinically as the injection formulation (Lianbizhi®) in China for the treatment of upper respiratory tract infections, bacillary dysentery, pneumonia and acute tonsillitis. Recent research suggested that ASB possessed a wide range of pharmacological activities such as a gastro-protective effect against indomethacin-induced gastric ulceration and competitive thiol groups for urease inhibition.

Andrographis paniculata Nees (family: Acanthaceae), commonly known as ‘king of bitters’, is a well-known medicinal herb found in India and is used in a variety of human illnesses. Andrographolide and 14-deoxy-11,12-didehydro-andrographolide are the two major bioactive constituents of this plant. Extracts of the plant and the major compounds have shown multiple pharmacological properties, such as antibacterial, antimalarial, immunostimulant, anti-HIV, anti-inflammatory, antihepatotoxic, anti-hyperglycemic, antimicrobial, cardiovascular, anti-diabetic and anticancer activities. Structurally, Andrographolide is a bicyclic diterpene with an α-alkylidene γ-butyrolactone ring, two olefin bonds, and three hydroxyls at C-3, C-14(secondary) and C-19 (primary), respectively. Structure activity relationship (SAR) studies of 1 showed that an intact γ-butyrolactone ring, double bonds at C-8 and C-12 and a hydroxyl group at C-14 are important for cytotoxic activity. Substitution of the acetyl functionality at the C-14 hydroxyl as well as protection of C-3 and C-19 hydroxyls with aromatic aldehydes increased the cytotoxicity. Also, the 15-alkylidene derivatives with the protection of hydroxyls at C-3 and C-19 by aromatic aldehydes were reported as potent α-glucosidase inhibitors.

Andrographis paniculata has been used in traditional medicine in Asian countries for centuries. Andrographolide, 1 and 14-deoxy-11,12-didehydroandrographolide 2 are major diterpene lactones with various biological potentials like analgesic, antipyretic, anti inflammatory antimicrobial, hepatoprotective, antiviral, antidiabetic and antitumor activities. Many of their synthetic analogues exhibit superior anticancer activity over the parent compounds in both in vitro and in vivo models. Interactions between , -unsaturated -butyrolactonemoieties, C-12,13 & C-8,17 double bonds, the C-14 hydroxyl or its ester moiety and protection of 3,19-hyroxyl with acetyl or suitable acetals/ketals are key for the cytotoxic activity exhibited by andrographolide and its analogues. Recent reviews also revealed that 1 and its derivatives are versatile bioactive molecules for combating inflammation and cancer.

Andrographis paniculata is an important herbal medicine traditionally used to treat a variety of diseases in some Southeast Asian countries such as China and India. Andrographolide, the main and most pharmacologically active diterpene lactone of the plant, has been reported to have anti-cancer, anti-inflammatory, antiviral, anti-allergic, anti-diabetic and immunostimulatory activities. For the improvement of antitumour effects, andrographolide was also utilized as a starting material for synthesis of derivatives. Some acylated andrographolide analogs gave good antitumour actions against various cancer cell lines. However, the selective monoacylation of andrographolide is difficult to achieve by means of conventional chemical approaches due to the similar reactivity of the hydroxyl groups in the molecule. Accordingly, the multi-step synthesis based on protection/deprotection reactions is required.
