1.Antidepressant-like effect of Casimiroa pubescens root extracts.

In this study, we investigated the antidepressant-like effects of the hexane (HCP), ethyl acetate (ECP) and methanol (MCP) extracts of the roots of Casimiroa pubescens Ramírez (Rutaceae) using the forced swim test (FST). In an initial experiment, each extract was orally administered to mice only once 60 min before to the FST. In a second experiment, doses were administered 24, 7 and 1 h before testing. Our results showed that the triple administration of the extracts provided a stronger effect than single administration. However, the combination of HCP at 7.5 mg/kg and imipramine (IMI) at 12.5 mg/kg showed the greatest effect. The coumarins 3-(1',1', dimethyl allyl)-herniarin, auraptene, 8-geranyl-oxy psoralen, isopimpinellin and the flavonoid zapotin were isolated from the extracts. The hexane extract of C. pubescens showed an antidepressant-like activity, which may inspire further studies on developing new antidepressant agents.
2.Capillary electrochromatography of selected phenolic compounds of Chamomilla recutita.

This article explores the use of capillary electrochromatography for the analysis of chamomile (Chamomilla recutita L.) extracts. After a thorough study of analytical parameters such as mobile and stationary phase composition, applied voltage, and temperature, a methodology to determine 11 bioactive phenolic compounds (coumarins: herniarin, umbelliferone; phenylpropanoids: chlorogenic acid, caffeic acid; flavones: apigenin, apigenin-7-O-glucoside, luteolin, luteolin-7-O-glucoside; flavonols: quercetin, rutin and flavanone: naringenin) in chamomile extracts was proposed. The method was performed in a Hypersil SCX/C18 column with pH 2.8 phosphate buffer at 50 mmol L(-1) containing 50% acetonitrile (pH adjusted before the addition of the organic solvent). All compounds were separated in less than 7.5 min under isocratic conditions. Figures of merit include linearity (peak area versus apigenin concentration) from 50.0-1000 microg/mL (r2=0.995), and intra-day precision of retention time and peak area better than 1.
3.Simultaneous determination of urinary metabolites of methoxypsoralens in human and Umbelliferae medicines by high-performance liquid chromatography.

A high-performance liquid chromatography method has been developed for the determination of coumarins and furocoumarins (psoralens). Nine coumarins and furocoumarins are separated simultaneously on a Hypersil C(8) (25 cm x 4.6-mm i.d.) column with a gradient of methanol and acetonitrile aqueous solution as mobile phase at 1.0 mL/min with two-channel UV-vis absorbance detection. The limits of detection are 0.366, 0.219, 0.317, 0.440, 0.536, 0.300, 0.531, 0.531, 0.237, and 0.280 ng/mL for coumarin, 7-hydroxycoumarin, 7-methoxycoumarin, citropten (5,7-dimethoxycoumarin), 7-ethoxy-4-methylcoumarin, psoralen, xanthotoxin (8-methoxypsoralen), bergapten (5-methoxypsoralen), isopimpinellin (5,8-dimethoxypsoralen), and imperatorin (9-isopenteneoxypsoralen), respectively. Human urine is analyzed 1-6 days after ingestion of the oral Chinese medicines. This lead to the conclusion that the concentration of coumarins and furocoumarins is higher than that of the control urine.
