ABT-751 (E-7010, Scheme 1) is an antitumor sulfonamide which inhibits tubulin polymerization by binding to tubulin at the colchicine binding site. It is currently in Phase II clinical study and has shown excellent antitumor activity against a variety of xenograft models, such as nonsmall cell lung cancer (NSCLC) and colon cancer. In addition, the crystal structure of ABT-751 has already been described. The title compounds, 4-(3-nitropyridin-2-ylamino) phenol (I) and 4-(3-aminopyridin-2-ylamino)phenol (II), are two intermediates for the synthesis of ABT-751. As shown in Scheme 1, the reaction of 4-aminophenol with 2-chloro-3-nitropyridine yielded I which was converted into II by reduction. Instead of the Pd/C catalytic hydrogenation described in literature, reduction with cheap sodium sulfide in aqueous media was utilized for shorting the reaction time and simplifying the operation. The crystal structures of the resultant compounds were determined by single-crystal X-ray diffraction.

With a view to studying biological activity of the new dithiophosphonates containing pharmacophoric phenol and pyrrole fragments, compound 3a was converted into ammonium salts. Unlike the initial hydrogen dithiophosphonate which is characterized by an unpleasant odor, its salts are almost odorless crystalline substances that are convenient to handle [1, 3]. Crystalline ammonium salt 4 was obtained by bubbling dry ammonia through a solution of 3a in benzene at 20℃ (Scheme 2).

Owing to the increasing stringency of requirements to the quality of phenol used for production of various polymers, including those exhibiting heat resistance, it is necessary to study in more detail processes of distillation of a mixture of phenol with concomitant impurities in the stage of phenol isolation as a commercial product. Among the main impurities determining the quality of commercial phenol are water, α-methylstyrene, and 2-methylbenzofuran, with the last compound being a product of reaction between hydroxyacetone and phenol in the final stages of phenol isolation.
